Oviduct EP with the NEPA21 – GONAD Technique

Oviduct EP with the NEPA21 – GONAD Technique
Please note page page 9 of the following article:
GONAD: Genome-editing via oviductal Nucleic Acids delivery system: a novel microinjection independent genome engineering method in mice
Dr. Masato Otsuka et al.
Nature Scientific Reports, June 2015. DOI:10.1038/srep11406

The article describes how to electroporate Oviducts in situ.
One of the article conclusions is that the EP conditions it utilised may require further refinement and optimisation – in order to delivered a gentler pulse.

Please note that the author Dr. Masato Otsuka, now uses our NEPA21 system to deliver the finer control he identified as missing in the above reference article.  He has obtained even better results than those reported in his publication.

For your further information, Dr. Masato Otsuka also uses our CUY52P2.5X4 electrode for his GONAD method.  The electrode surface is concaved (bowl-like shape) and thus makes it easier to grasp/hold the oviduct. The electrode is to Dr. Ohtsuka’s specific design.  He requested that we manufacture it for him.  With this electrode, he is obtaining better results than with the previous electrode configuration he used

The NEPA21 both uniquely monitors in real-time the delivered energy (Joules) and enables the researcher to matches the energy required for the different ‘poration’ and ‘payload driving’ events.  (Please email sales@sonidel.com to request a synopsis of how the Poration Pulse enhances Viability and how the Transfer Pulse delivers a gentler pulse to drive the payload firstly in the direction of the current and then in the opposite direction, via polarity switching).

The Otsuka article above also referenced the Kaneko article “Simple knockout by electroporation of engineered endonucleases into intact rat embryos”.
The Kaneko article also uses our NEPA21 system.
