Mouse/Rat - Skin
Electrode Recommendation:
To deliver genes into Mouse/Rat Skin by electroporation, we recommend:
- CUY663-5×10 Technical Drawing: CUY663 series
This electrode is a tweezers-type electrode. One tweezers is a 5 x 10mm rectangular electrode and the other is three needles which look like a fork.
For your further information, some clients prefer to use the CUY650P electrode series for the same application. The CUY650P series is also a tweezers type electrode but it incorporates disk electrodes. With the CUY650P series, skin is picked up and simply sandwiched between the disk electrodes.
Technical Drawing: CUY650 series
Protocol Recommendation:
Kindly note Dr. Maruyama’s article:
- ‘Epidermis-targeted gene transfer using in vivo electroporation’.
In this article Dr. Maruyama uses the CUY21EDIT electroporator and CUY663B (currently catalogue number CUY663-5×10) electrode.
According to Dr. Maruyama’s paper, the required voltage is maximum 24V. The appropriate electrode diameter depends on the size of the area into which the genes are to be transfected. If one’s target area is small, the 1mm diameter (CUY650P1) electrode is appropriate.
