Plant Protoplast Fusion By Electro Cell Fusion

Electro Cell Fusion Protocol:

Plant Protoplast Fusion may be performed by Electro Cell Fusion:

–  ‘Plant Protoplast Fusion by Electro Cell Fusion’

(Cell Type: General Plant Protoplast and Celery Apium Graveolens L. Fusio)

The stated methodologies require the use of the cell fusion generator LF201 or LF101 and a chamber.

LF101 LF201

Electro Cell Fusion Electrode Recommendation:

Per ‘Plant Protoplast Fusion by Electro Cell Fusion’, the most appropriate electrodes for plant protoplast fusion, are:

–  CUY501G4(160ul) Technical Drawing: CUY501G

–  CUY498-4(2.0ml) and             Technical Drawing: CUY494-4

–  CUY498-10(4.0ml) Technical Drawing: CUY498-10

The number in brackets indicates the volume between electrodes.  If one has a preference for the larger volume chamber electrode, then we would recommend the CUY498-10 electrode.

Further electrode information:

1.  Our electrodes for LF101 and LF201 are compatible with BTX ECM2001. However, they cannot be used with the Eppendorf device.

2.  If using our electrodes with another device, both the C115 and C117 are required.

3.  Unlike the CUY21 range of electroporators, the LF101 and LF201 do not have an automatic safety cut-off switch. As compared with that of CUY21 range, the DC pulse length of both the LF101 and LF201 is very short (usec).  In addition, unlike the electrodes used in in vivo electroporation, the researcher does not need to hold the electrodes used in cell fusion, so there is no need for an automatic safety cut-off switch in the LF101 and LF201.
